Not only was SOD activity universally present in oxygen-metabolizing organisms, its concentration was found in a remarkably narrow range, whether in human brain or a tomato or Escherichia coli. This implied two things: that the metabolism of oxygen inevitably leads to superoxide production, and that the free radical, if left unscavenged, must be seriously toxic.

The great advantage of the SOD catalyzed over the spontaneous dismutation is due to several factors. First, the rate constant of the enzyme-catalyzed reaction exceeds that of the spontaneous reaction, secondly the steady-state concentration of the O2•− is much less than the average concentration of the SOD; hence, an O2•− is far more likely to collide with SOD than it is to collide with another O2•− or HO2•, and thirdly the spontaneous reaction is second order in O2•−/HO2•, while the catalyzed reaction is first order with respect to O2•−.
